Yes it requires a gasket, usually included with the pump. Please reference the factory service manual for the proper procedure.

When you remove the water pump most of the coolant will come out. Might as well flush now and fill with Toyota red coolant.

I'm going to guess you are quite new to working on cars. One thing you need to learn is to do one thing at a time and not pull out the parts cannon. You have done the water pump and now the car leaks and doesn't start. Solve those issues first - a bad thermostat or electric fan wont cause starting issues.
